USPIO-PEG-sLeX is a nanotheranostic agent composed of ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles coated with polyethylene glycol (PEG) and conjugated to Sialyl Lewis X (sLeX), a carbohydrate ligand. This multifunctional nanoparticle is designed for targeted imaging and therapy in oncology. The sLeX moiety enables specific targeting of E-selectin, which is overexpressed on tumor vasculature and metastatic sites. The USPIO core provides T2-weighted contrast enhancement for magnetic resonance imaging (MRI), allowing non-invasive visualization of tumors and metastases[2][3][4]. Additionally, the agent exhibits photothermal properties; upon irradiation, it can induce localized hyperthermia that enhances immune cell infiltration into tumors and amplifies the efficacy of immune checkpoint blockade therapies such as anti-PD-L1 antibodies[1]. This dual functionality positions USPIO-PEG-sLeX as both a diagnostic probe for MRI-based detection/quantification of E-selectin expression in tumors and a therapeutic enhancer in cancer immunotherapy.
