ustekinumab + abatacept

ustekinumab + abatacept is a sequential immunotherapy regimen investigated for the treatment of moderate to severe plaque psoriasis. The regimen consists of an induction phase using **ustekinumab**, a human monoclonal antibody that inhibits the p40 subunit shared by interleukins **IL-12** and **IL-23**, followed by a maintenance phase using **abatacept**, a recombinant fusion protein (CTLA-4-Ig) that blocks T-cell costimulation by binding to **CD80** and **CD86**. This approach was evaluated in the Phase 2 **PAUSE** trial (ITN059AI), sponsored by the **National Institute of Allergy and Infectious Diseases (NIAID)** and the Immune Tolerance Network, to determine if initial cytokine blockade followed by T-cell modulation could induce long-term immune tolerance and prevent disease relapse in patients who have responded to induction therapy.
