UTA2-1-loaded PD-L-silenced dendritic cells is an investigational cell-based biological therapy developed by VU University Medical Center. The therapy consists of donor-derived monocyte-derived dendritic cells (DCs) that are loaded with the minor histocompatibility antigen (MiHA) UTA2-1, a B-cell lineage-restricted antigen. To enhance the potency of the vaccine, the expression of immune checkpoint ligands PD-L1 and PD-L2 is silenced on the dendritic cells using small interfering RNA (siRNA), thereby removing a key inhibitory brake on T-cell activation. This vaccine is primarily evaluated in the context of allogeneic stem cell transplantation for B-cell hematological malignancies, including multiple myeloma and chronic lymphocytic leukemia, where it is administered in combination with donor lymphocyte infusion (DLI) to augment graft-versus-tumor (GVT) activity.
