UTA2-1 peptide-loaded PD-L-silenced donor dendritic cell vaccine is an investigational, donor-derived dendritic cell (DC)-based immunotherapy developed by Vrije Universiteit Medisch Centrum (VUmc). The vaccine consists of donor-derived dendritic cells loaded with peptides of the minor histocompatibility antigen (MiHA) UTA2-1, in which the expression of programmed death ligands 1 and 2 (PD-L1 and PD-L2) has been silenced using small interfering RNA (siRNA). This silencing is designed to prevent T-cell inhibition and enhance the immunogenicity of the vaccine. It was investigated for the treatment of B-cell hematologic malignancies, including multiple myeloma, chronic lymphocytic leukemia (CLL), and non-Hodgkin lymphoma, specifically in patients with residual or relapsed disease following allogeneic stem cell transplantation (alloSCT) who failed a first donor lymphocyte infusion (DLI). The vaccine was intended to be administered in combination with a second DLI to boost graft-versus-tumor (GVT) activity while minimizing graft-versus-host disease (GvHD). However, the clinical development of this vaccine has been terminated.
