Overview

UTR-HER2-mBCa is an investigational RNA-based therapeutic developed by UTR Therapeutics for the treatment of metastatic HER2-positive (HER2+) trastuzumab-resistant breast cancer. The therapeutic utilizes a proprietary technology involving engineered destabilized 3' untranslated region (3'UTR) AU-rich elements (ARE) to selectively degrade HER2/ERBB2 mRNA transcripts. By replacing the naturally stable poly-U sequences in the 3'UTR of the ERBB2 gene with these destabilized sequences, the drug triggers the rapid degradation of the mRNA, leading to a significant reduction in HER2 protein expression. This mechanism is designed to overcome resistance mechanisms encountered with traditional HER2-targeted therapies, such as monoclonal antibodies or tyrosine kinase inhibitors, and is currently in the IND-enabling stage of development.
