Overview

UV-4B is an orally administered small molecule iminosugar antiviral drug candidate. It acts as a competitive inhibitor of host endoplasmic reticulum α-glucosidase I and II enzymes (alpha-glucosidase inhibitors), disrupting glycan processing and folding of viral glycoproteins. This leads to misfolded viral proteins, impairing virus assembly and secretion, thereby reducing the infectivity of viruses such as dengue virus (all four serotypes), influenza A and B viruses, and filoviruses. The drug has demonstrated in vitro activity against these viruses and in vivo efficacy in animal models for dengue and influenza. Clinical development has included phase 1 studies for safety in healthy volunteers; it was discontinued for influenza but remains under investigation for dengue fever[1][3][5][6][8].
