Overview

**Vγ4Vδ5 TCR-anti CD3 bispecific engager** is an experimental bispecific T-cell engager (TCE) constructed by fusing the extracellular domains of a tumor-reactive Vγ4Vδ5 T-cell receptor (TCR) to an anti-CD3 single-chain variable fragment (scFv). It redirects conventional αβ T cells to tumor cells by simultaneously binding CD3 on T cells and tumor-associated antigens recognized by the γδ TCR, mimicking the phosphoantigen-dependent recognition involving BTN2A1/BTN3A1 on cancer cells. Developed as part of gamma delta TCR anti-CD3 bispecific molecules (GABs), it demonstrates potent in vitro and in vivo antitumor activity against various tumor lines and patient-derived material, with the γ-TCR HV4 loop critical for target engagement, while avoiding direct activation of endogenous γδ T cells.[1][4][6]
