Overview

vδ2 T cells are a specific subset of gamma-delta (γδ) T cells that possess innate-like anti-tumor properties, primarily recognizing phosphoantigens presented by butyrophilin (BTN) family proteins (such as BTN3A1 and BTN2A1) on the surface of malignant cells. In the context of the program developed by the Chinese PLA General Hospital, these cells are utilized as an allogeneic, off-the-shelf cell therapy. The product is genetically engineered to be TCR-enhanced by incorporating a specific BTN protein-binding moiety on the cell surface, which significantly boosts the cells' intrinsic tumor-killing potency. To enhance in vivo persistence and prevent T-cell exhaustion, the engineered cells are designed without traditional co-stimulatory signaling domains or CD3ζ domains. This therapeutic approach is currently being investigated in Phase I/II clinical trials for patients with relapsed or refractory hematologic malignancies and advanced solid tumors.
