V-scVLPs are virus-like silicon nanoparticle vaccines designed for efficient co-delivery of hepatocellular carcinoma (HCC)-specific neoantigens and a Toll-like receptor 9 (TLR9) agonist to dendritic cells via caveolin-mediated endocytosis. This intracellular delivery enhances DC maturation, primes robust CD8+ T cell responses, and central memory T cell responses to prevent tumor growth. In established tumor models, V-scVLPs upregulate TIM-3 in CD8+ T cells; combining with TIM-3 blockade further improves antitumor activity by reducing regulatory T cells and increasing cytokines for improved immunotherapy outcomes in HCC animal models[1][2][4][7].
