v128-EGFP is an engineered adeno-associated virus (AAV) vector designed for high-efficiency gene delivery to pancreatic alpha cells. It utilizes a novel AAV8 variant capsid, created through directed evolution and modification of Variable Surface Region IV, which demonstrates significantly enhanced tropism for human pancreatic alpha cells compared to wild-type AAV8. The vector carries the Enhanced Green Fluorescent Protein (EGFP) as a reporter transgene. Developed by Chengdu Origen Biotechnology and partners, this platform is intended to overcome historical barriers in islet transduction, providing a foundation for next-generation gene therapies for metabolic diseases such as Type 1 Diabetes, potentially enabling "Artificial Pancreas" approaches.
