Melevodopa is a prodrug of levodopa (L-dopa), acting as the precursor of dopamine, while carbidopa inhibits aromatic L-amino acid decarboxylase, preventing peripheral breakdown of levodopa and increasing its availability to the brain. Entacapone is a selective, reversible inhibitor of catechol-O-methyltransferase (COMT), preventing the O-methylation and further breakdown of levodopa in the periphery, thereby enhancing plasma levodopa levels and prolonging its therapeutic effects. This combination is developed to offer improved and more consistent pharmacokinetics for levodopa delivery compared to standard oral L-dopa/carbidopa, aiming for reduced motor fluctuations in patients with **Parkinson's disease**.
