V3-750 is a pH-sensitive peptide-based imaging probe designed for the detection and intraoperative visualization of pancreatic ductal adenocarcinoma (PDAC). It consists of a pH low insertion peptide (pHLIP), specifically the V3 variant, conjugated to the near-infrared fluorescent dye HiLyte 750. The probe targets the acidic microenvironment characteristic of solid tumors; at low pH (typically 6.2–6.8 in pancreatic tumors), the peptide undergoes a conformational change that allows it to insert into the cell membrane, thereby localizing the fluorescent signal to the tumor site. This mechanism enables high-contrast imaging using multispectral optoacoustic tomography (MSOT) and near-infrared fluorescence (NIRF) imaging. Developed by researchers at the University of Oklahoma and the Military University Hospital Prague, V3-750 is intended to improve the identification of tumor margins and molecular features during surgery.
