V4020 is a live-attenuated vaccine candidate designed to protect against Venezuelan Equine Encephalitis Virus (VEEV). Developed through a collaboration involving the U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID) and Medigen Biotechnology, the vaccine is derived from the virulent Trinidad Donkey (TrD) strain of VEEV. It incorporates three specific attenuating mutations—one in the 5' untranslated region (UTR) and two in the E2 and E1 envelope glycoproteins—to enhance safety and reduce the risk of reversion to virulence compared to earlier vaccine candidates like TC-83. V4020 is intended for the prevention of VEEV infection, a significant biothreat and zoonotic pathogen, and has been evaluated in Phase 1 clinical trials for safety and immunogenicity via subcutaneous and intramuscular administration.
