Drug intelligence / Profile preview
VAC is a chemotherapy combination comprising **vincristine**, **dactinomycin** (also called Actinomycin D), and **cyclophosphamide**. It is primarily used in the treatment of various types of **sarcomas**, particularly in children. The combination is administered **intravenously**, typically via a central line, PICC line, or portacath. VAC is commonly used for conditions such as **Ewing sarcoma**, **soft tissue sarcomas** (including desmoplastic small round cell tumour and rhabdomyosarcoma), and **desmoid-type fibromatosis** (desmoid tumour). For Ewing sarcoma, rhabdomyosarcoma, and desmoid-type fibromatosis, treatment often involves up to 7 cycles, lasting approximately 5 months. For desmoplastic small round cell tumour, VAC is continued as long as it is effective and side effects are manageable. The mechanism of action involves **destroying rapidly dividing cells**, such as cancer cells. Each component drug acts differently to inhibit or kill tumor cells. Clinical trials have evaluated VAC in comparison to other regimens (e.g., VAC/VTC for intermediate-risk rhabdomyosarcoma) and studied its use with or without radiation therapy in specific sarcoma types. Potential side effects include changes in liver function, issues with sperm production, chills, weakness, numbness or tingling (neuropathy), and nerve damage causing pain or loss of sensation. The combination is approved by the FDA for treating cancer or related conditions, specifically **ovarian germ cell tumors** and **rhabdomyosarcoma in children**.
