**vaccine-Centre Hospitalier Universitaire Vaudois-melanoma** is an investigational multipeptide therapeutic cancer-vaccine regimen developed at Lausanne University Hospital Center for HLA-A2-positive stage II-IV melanoma. The subcutaneously administered formulation combines five synthetic tumor-antigen peptides, recombinant LAG-3Ig fusion protein eftilagimod alpha, and Montanide ISA-51 adjuvant. Four short 9/10-mer peptides are intended to stimulate CD8-positive T cells, while one 15-mer peptide is intended to stimulate CD4-positive T cells; the peptide antigens derive from MAGE-A3, NY-ESO-1, and Melan-A. Eftilagimod alpha binds major histocompatibility complex class II molecules on antigen-presenting cells and promotes antigen-presenting-cell and downstream T-cell activation. In the Phase I/IIa study NCT01308294, 16 metastatic melanoma patients received up to nine vaccinations, with antigen-specific CD4-positive and CD8-positive T-cell responses and favorable tolerability reported.
