Vaccinia-tyrosinase is a therapeutic cancer vaccine consisting of a recombinant vaccinia virus engineered to express the full-length human tyrosinase gene. Tyrosinase is an enzyme essential for melanin synthesis and is commonly overexpressed in melanoma cells. The vaccine aims to induce an immune response against tyrosinase-expressing tumor cells by presenting the antigen via viral infection of host cells. This approach leverages the immunogenicity of both the viral vector and the tumor-associated antigen (tyrosinase) to stimulate cytotoxic T lymphocyte responses targeting melanoma. Clinical studies have evaluated this strategy in patients with metastatic melanoma, often in combination with interleukin-2 (IL-2), showing some evidence of immune activation and partial clinical responses[1][2][8]. The National Cancer Institute has been a primary developer and sponsor for clinical trials involving this agent.
