Overview

Vactosertib + imatinib is an investigational drug combination being developed by MedPacto for the treatment of desmoid tumors (aggressive fibromatosis). Vactosertib (also known as TEW-7197) is an orally bioavailable small molecule inhibitor of the TGF-β type I receptor kinase (also called activin receptor-like kinase 5 or ALK5). Imatinib is a well-established tyrosine kinase inhibitor (TKI) that targets BCR-ABL, c-KIT, and PDGFR. The combination is designed to provide synergistic antitumor activity by simultaneously inhibiting the TGF-β signaling pathway, which is often implicated in tumor progression and fibrosis, and tyrosine kinase-mediated growth signals. Clinical data from Phase Ib/II trials have demonstrated promising activity in patients with progressive, locally advanced desmoid tumors, showing high progression-free rates at 16 weeks and 1 year.
