Overview

Sprint Bioscience is developing a small molecule inhibitor of Vaccinia-Related Kinase 1 (VRK1) for the treatment of solid tumors, specifically targeting glioblastoma and neuroblastoma. The program, designated VADA, leverages the concept of synthetic lethality; VRK1 inhibition is selectively toxic to cancer cells that have low expression of the related kinase VRK2, a genetic profile found in the majority of high-grade gliomas and high-risk neuroblastomas. VRK1 is a key regulator of the DNA damage response and cell cycle progression. The program was briefly licensed to Day One Biopharmaceuticals before being returned to Sprint Bioscience in 2025. It is currently in the preclinical stage of development.
