VADC45 is a preclinical antibody-drug conjugate (ADC) developed by Vor Bio targeting the CD45 protein, which is broadly expressed on hematopoietic stem and progenitor cells (HSPCs) as well as mature myeloid and lymphoid immune cells. The drug consists of a human IgG1 antibody linked to the DGN549-C linker-payload, a clinically validated cytotoxic payload used in other ADCs. VADC45 is engineered for high specificity to human CD45 with a short half-life, aiming to selectively deplete hematopoietic and immune cells while sparing shielded or modified stem cells. Its primary intended use is as a targeted conditioning agent prior to allogeneic hematopoietic cell transplant (HCT), gene therapies (such as for sickle cell disease), or for holistic immune reset in autoimmune disorders. By targeting CD45, VADC45 seeks to improve safety and tolerability compared with traditional conditioning regimens, reduce allograft rejection risk, and enable new therapeutic strategies such as epitope modification of CD45 on healthy donor HSPCs[1][3][4][6].
