Vadimezan + paclitaxel is a combination regimen used in clinical research, primarily for the treatment of advanced non-small cell lung cancer. Vadimezan (code names ASA404, DMXAA) is a small molecule tumor-vascular disrupting agent (Tumor VDA) that acts by causing irreversible disruption of tumor vasculature and inducing hemorrhagic necrosis within the tumor core. Paclitaxel is a cytotoxic small molecule that stabilizes microtubules and inhibits cell division. Combination therapy was investigated in phase I-III trials; however, in large randomized phase III studies the addition of vadimezan to standard chemotherapy (including paclitaxel) failed to improve efficacy over paclitaxel-containing regimens alone, and development was discontinued.
