Overview

Vaginal nifedipine ointment is a compounded topical formulation containing 0.2% nifedipine in a lanolin and mineral oil base. Developed by Vanderbilt University, it was investigated as an adjunctive treatment to pelvic floor physical therapy for female patients suffering from levator ani syndrome (also known as levator myalgia) and pelvic floor hypertonus. Nifedipine is a dihydropyridine calcium channel blocker that acts by inhibiting the influx of calcium ions into smooth and skeletal muscle cells, thereby promoting muscle relaxation and reducing associated pelvic pain, dyspareunia, and dysfunctional voiding. A randomized controlled trial (NCT01586286) was designed to evaluate its efficacy, but the study was ultimately withdrawn before enrollment began.
