**ValloVax** is a therapeutic cancer vaccine derived from human placental endothelial cells treated with interferon-gamma (IFN-γ) to upregulate HLA and costimulatory molecules (CD40, CD80, CD86), enhancing immunogenicity. It induces tissue-nonspecific antitumor immunity by stimulating T cell and B cell responses that produce IgG antibodies targeting tumor endothelial cell antigens such as VEGFR1, VEGFR2, TEM1 (endosialin), CD105 (endoglin), ROBO4, FGFR2, and EGFR, leading to selective killing of tumor vasculature via apoptosis, reduced tumor oxygenation, and tumor regression. Developed by Batu Biologics, it has shown efficacy in preclinical models of lung cancer, melanoma, breast cancer, colon cancer, glioma, and glioblastoma, with synergy when combined with PD-1 or CTLA-4 checkpoint inhibitors; it received FDA IND clearance for Phase 1 trials in solid tumors including non-small cell lung cancer and glioblastoma, and demonstrated safety in a pilot human trial.[1][2][5][6][14]
