VAN-305 is a biologic drug candidate being developed by Valin Technologies for the treatment of intracranial hemorrhage (ICH) and other bleeding disorders. It is a recombinant, catalytically inactive mutant of tissue plasminogen activator (tPA) containing an S481A mutation. This specific mutation renders the protein enzymatically inert while allowing it to function as a competitive inhibitor of endogenous human tPA and urokinase-type plasminogen activator (uPA). By competing for binding sites and substrates within the fibrinolytic system, VAN-305 effectively suppresses the breakdown of fibrin, thereby stabilizing blood clots and reducing hematoma expansion in patients with hemorrhagic stroke. The drug is currently in Phase 1 clinical development, with Sinopharm acting as a strategic partner for its development in China.
