Overview

vancomycin + tobramycin is an investigational antibiotic combination being studied as an impregnated bone graft intervention in the ABOGRAFT clinical trial. Developed by University Hospital Linköping, the treatment involves morselized bone allograft loaded with 1 gram of vancomycin powder and 8 ml of liquid tobramycin. This combination is intended for local administration during hip arthroplasty (hip replacement) to prevent periprosthetic joint infections (PJI). Vancomycin is a glycopeptide antibiotic that inhibits bacterial cell wall synthesis in Gram-positive bacteria, while tobramycin is an aminoglycoside that inhibits protein synthesis in Gram-negative bacteria by binding to the 30S ribosomal subunit. The intervention is currently being evaluated in a randomized, double-blind, placebo-controlled trial to determine its efficacy in reducing re-operation rates due to infection.
