vandetanib + bicalutamide

**vandetanib + bicalutamide** is a fixed-dose combination of two oral small molecule drugs: vandetanib, a multi-targeted tyrosine kinase inhibitor that inhibits vascular endothelial growth factor receptor (VEGFR), epidermal growth factor receptor (EGFR), and RET kinases; and bicalutamide, a nonsteroidal antiandrogen that acts as an androgen receptor antagonist. The combination was investigated for treatment of metastatic castration-resistant prostate cancer (mCRPC), aiming to simultaneously block angiogenic, proliferative, and androgen-signaling pathways. In phase II trials, the combination did not show efficacy superior to bicalutamide alone and was associated with increased toxicity, leading to discontinuation of further development for this indication[1][2].
