Overview

**vandetanib + FOLFOX** is an investigational combination regimen consisting of **vandetanib**, a small molecule tyrosine kinase inhibitor, and **FOLFOX**, a standard chemotherapeutic regimen used primarily in colorectal and other gastrointestinal cancers. - Vandetanib inhibits multiple signaling pathways, including vascular endothelial growth factor receptor (VEGFR), epidermal growth factor receptor (EGFR), and RET tyrosine kinases, exerting both antineoplastic and antiangiogenic activity. - FOLFOX is a combination of three drugs: folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin. - Fluorouracil is an antimetabolite that interferes with DNA synthesis in rapidly dividing cells. - Oxaliplatin is a platinum-based agent that forms DNA adducts causing cell death. - Folinic acid enhances the effectiveness of fluorouracil. This combination aims to synergize the molecular targeting of vandetanib with the cytotoxic effects of FOLFOX to treat advanced cancers, most commonly in the setting of metastatic colorectal or gastrointestinal malignancies.
