Drug intelligence / Profile preview
VANL-100 is a novel synthetic antioxidant co-drug developed by covalently linking naringenin (a naturally occurring flavanone) and alpha-lipoic acid (a potent mitochondrial antioxidant) via an ester bond. Developed by researchers at the University of Guelph, VANL-100 is designed to target oxidative stress, amyloid-beta (Aβ) accumulation, and tau hyperphosphorylation, which are key pathological hallmarks of Alzheimer's disease and ischemic stroke. In preclinical models, VANL-100 has demonstrated significant neuroprotective and cytoprotective properties, showing up to 100- to 10,000-fold greater potency than its parent compounds alone or in combination. Mechanistically, VANL-100 modulates the Akt/GSK3β signaling pathway, inhibits glycogen synthase kinase-3β (GSK3β) activity, reduces tau phosphorylation, and increases ADAM10 protein expression to promote the non-amyloidogenic pathway of amyloid precursor protein (APP) processing.
