Vantictumab + docetaxel is an investigational combination therapy being studied primarily for the treatment of advanced solid tumors, notably non-small cell lung cancer (NSCLC). Vantictumab is a first-in-class monoclonal antibody that targets the Wnt signaling pathway, specifically acting as an antagonist of Frizzled family receptors, which are implicated in the self-renewal and maintenance of cancer stem cells. Docetaxel is a standard-of-care antineoplastic agent classified as a taxane, which acts by stabilizing microtubules and inhibiting cell division. Preclinical and early clinical data suggest this combination exhibits synergistic antitumor activity, with optimal effects seen when vantictumab is administered prior to docetaxel. The combination is being developed by OncoMed Pharmaceuticals (in collaboration with Bayer for some studies) and assessed in clinical trials for safety, pharmacokinetics, and preliminary efficacy, mainly in recurrent or advanced NSCLC and other solid tumors[1][2][3][4][5].
