This combination comprises **vantictumab** (a fully human monoclonal antibody and Wnt signaling pathway inhibitor that targets the Frizzled family receptors FZD1, FZD2, FZD5, FZD7, and FZD8), **nab-paclitaxel** (an albumin-bound formulation of paclitaxel), and **gemcitabine** (a nucleoside analog used as chemotherapy)[3][4]. It has been investigated in phase 1b studies for the treatment of untreated metastatic pancreatic adenocarcinoma, aiming to augment cytotoxic chemotherapy by also targeting cancer stem cell pathways (through Wnt inhibition)[1][3][4]. Early clinical results showed partial responses and disease stabilization in some patients, but development was halted due to bone-related adverse events linked to vantictumab[3][4]. Vantictumab was developed by OncoMed Pharmaceuticals and the combination was tested in studies sponsored by OncoMed Pharmaceuticals and later, Mereo BioPharma[4][5].
