Overview

Vapendavir is an orally active small molecule antiviral drug that acts as a capsid-binding inhibitor targeting human rhinoviruses (HRVs), the viruses responsible for the common cold and significant respiratory complications in patients with asthma and chronic obstructive pulmonary disease (COPD). It binds tightly to the VP1 protein canyon on the viral capsid, disrupting its ability to attach to cell surface receptors and inhibiting viral RNA release during uncoating, thereby blocking viral replication. Vapendavir has demonstrated broad-spectrum activity against approximately 97% of rhinoviruses tested. It is designed for oral administration and has shown favorable pharmacokinetics including high bioavailability and linear profile unaffected by concomitant medications. The drug is currently in Phase II clinical trials primarily for treatment of rhinovirus-induced exacerbations of asthma and COPD. Vapendavir shares a similar mechanism of action with lenacapavir, an FDA-approved HIV capsid inhibitor marketed by Gilead Sciences. The development has been led by companies including Biota Pharmaceuticals, Aviragen Therapeutics (now Vaxart), Altesa BioSciences among others[1][2][3][6][7][9].
