A clinical research program led by the University of California, San Diego (UCSD) investigating the use of vaporized cannabis flower for the acute treatment of migraine. The research evaluates different potencies and ratios of delta-9-tetrahydrocannabinol (THC) and cannabidiol (CBD), including THC-dominant (up to 10% THC), CBD-dominant, and combination formulations. The mechanism of action is believed to involve the activation of cannabinoid receptors (CB1 and CB2), which in turn inhibits the release of calcitonin gene-related peptide (CGRP), reduces CGRP-induced nitric oxide release, and suppresses trigeminovascular neurons and cortical spreading depression. The study was conducted as a randomized, double-blind, placebo-controlled crossover trial, with the combination formulation showing superior efficacy compared to placebo.
