VAR-102 is an oral small molecule inhibitor targeting atypical protein kinase C iota (aPKCi), a member of the protein kinase C family implicated as an oncogene in various human cancers. The drug is being developed for the treatment of a broad range of solid tumors, including non-small cell lung cancer (NSCLC), pancreatic cancer, and colorectal cancer. Preclinical studies have demonstrated that inhibition of aPKCi by VAR-102 leads to dose-dependent anti-tumor activity in both murine and human basal cell carcinoma (BCC) cell lines as well as other cancer models. Mechanistically, aPKCi plays a central role in regulating key oncogenic transcription factors such as GLI-1 and K-RAS, which are involved in multiple signaling pathways including the Hedgehog pathway. By inhibiting aPKCi, VAR-102 may modulate tumor growth and enhance immune recognition of tumors[1][4][5][9].
