Drug intelligence / Profile preview
VAR2CSA peptide-decorated antigenic liposomes is a preclinical multivalent peptide vaccine candidate being developed by the University of Alberta for the prevention of placental malaria. The vaccine consists of liposomes decorated with two conserved peptides, P6 (located in the DBL3X domain) and P10 (located in the DBL4epsilon domain), which map to conserved epitopes on the Plasmodium falciparum virulence factor VAR2CSA. These peptides are conjugated to a lipid linker via strain-promoted azide-alkyne cycloaddition chemistry. The liposomes encapsulate ovalbumin to enhance immunogenicity and incorporate the TLR4 agonist monophosphoryl lipid A (MPLA) as an adjuvant. The vaccine is designed to induce synergistic inhibitory antibodies that prevent the binding of infected red blood cells to chondroitin sulfate A (CSA) in the placenta.
