Drug intelligence / Profile preview
VAR2CSA vaccine refers to a class of placental malaria vaccine candidates designed to elicit protective antibodies against VAR2CSA, a member of the Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) family. VAR2CSA is expressed on the surface of infected erythrocytes and mediates their sequestration in the placenta by binding to the host receptor chondroitin sulfate A (CSA), leading to severe pregnancy outcomes such as maternal anemia, low birth weight, and stillbirth. Leading VAR2CSA-based vaccine candidates include PRIMVAC (developed by Inserm) and PAMVAC (developed by the University of Copenhagen), both of which have completed Phase I clinical trials. Next-generation candidates, such as HPISVpmv1, utilize structure-guided design to improve stability, yield, and the breadth of the immune response against polymorphic parasite strains.
