RT234 is an investigational dry-powder inhaled formulation of vardenafil developed by Respira Therapeutics for the treatment of pulmonary arterial hypertension (PAH). Vardenafil is a potent phosphodiesterase type 5 (PDE5) inhibitor that acts as a vasodilator. Unlike oral PDE5 inhibitors approved for erectile dysfunction and chronic PAH therapy, RT234 is designed for as-needed (PRN) use to acutely improve exercise tolerance and reduce exertional symptoms in PAH patients. The drug-device combination uses Respira’s proprietary Axial Oscillating Sphere Dry Powder Inhaler technology to maximize drug delivery to the distal lung. It has received Orphan Drug designation from the FDA for PAH and has demonstrated safety and tolerability in early-phase clinical studies[1][2][3][4][5][7].
