Varespladib + marimastat is an oral small molecule combination therapy being developed by Ophirex for the treatment of snakebite envenomation. The combination is designed to provide broad-spectrum protection against two major classes of snake venom toxins: secretory phospholipase A2 (sPLA2) and snake venom metalloproteinases (SVMPs). Varespladib acts as a potent inhibitor of sPLA2, which is responsible for neurotoxicity, local tissue injury, and hemorrhage, while marimastat inhibits SVMPs, which play a primary role in tissue digestion, necrosis, and coagulopathy. By combining these two complementary inhibitors, the therapy aims to rapidly mitigate toxin activity and prevent severe systemic and local pathologies in the field before hospital-based antivenom can be administered.
