Varlilumab + atezolizumab

Varlilumab + atezolizumab is an investigational combination therapy for cancer immunotherapy, pairing two monoclonal antibodies with complementary mechanisms of action. Varlilumab is a potent anti-CD27 agonist that activates and proliferates human T cells by stimulating the CD27 receptor, thereby enhancing immune responses against tumors. Atezolizumab is a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and B7.1 on T cells to prevent tumor-induced immune suppression[6][3]. The combination aims to synergistically activate anti-tumor T cell responses by both promoting T cell activation (via CD27) and removing inhibitory signals (via PD-L1 blockade). This approach has shown promising activity in early-phase clinical trials, particularly in renal cell carcinoma[8][6].
