Overview

Varlilumab + sunitinib is an investigational combination therapy being studied primarily for cancer treatment, notably in advanced or refractory clear cell renal cell carcinoma (CC-RCC) and potentially other solid tumors. Varlilumab is a fully human monoclonal agonist antibody that targets and activates CD27, a co-stimulatory molecule involved in T-cell activation, thereby enhancing anti-tumor immune responses. Sunitinib is an orally administered small-molecule multi-targeted receptor tyrosine kinase inhibitor that blocks several kinases implicated in tumor growth, angiogenesis, and metastasis—including platelet-derived growth factor receptors (PDGFRs), vascular endothelial growth factor receptors (VEGFRs), KIT (CD117), RET, CSF-1R, and FLT3. The rationale for combining these agents lies in their complementary mechanisms: sunitinib can modulate the tumor microenvironment to reduce immune suppression while varlilumab stimulates T-cell mediated immunity against tumors[5][8]. This combination has shown promising immunologic activity and clinical responses in early-phase studies.
