VAS2870 is a pan-NADPH oxidase (NOX) inhibitor, structurally identified as 3-benzyl-7-(2-benzoxazolyl)thio-1,2,3-triazolo[4,5-d]pyrimidine. It is widely used as a research tool to suppress NOX1, NOX2, and NOX4-mediated reactive oxygen species (ROS) production in various cell types, but does not inhibit NOX5. VAS2870 attenuates platelet activation, endothelial calcium signaling, migration, proliferation, and reduces oxidative stress via inhibition of ROS-generating NOX enzymes. It has shown efficacy in cell and animal models of cardiovascular disease, stroke, and fibrosis. VAS2870 works through noncompetitive inhibition of NOX isoforms and demonstrates limited off-target effects, showing no significant interaction with other oxidase systems or direct antioxidant activity[1][3][6].
