Vascular endothelial growth factor C (VEGF-C) is a secreted protein growth factor in the platelet-derived growth factor/vascular endothelial growth factor (PDGF/VEGF) family, encoded by the VEGFC gene. VEGF-C promotes lymphangiogenesis (growth of lymphatic vessels) by binding and activating its primary receptor, VEGFR-3 (FLT4), on lymphatic endothelial cells, stimulating their survival, proliferation, and migration. It also can bind and activate VEGFR-2, affecting blood vessel permeability and angiogenesis. VEGF-C is essential for lymphatic system development and maintenance in mammals. Therapeutically, VEGF-C is under development and investigation, notably as Lymfactin for lymphedema treatment, and is being explored in gene therapy for augmenting lymphangiogenesis in lymphatic disorders. Overproduction is linked to tumor lymphangiogenesis and metastasis; hence, it is also a relevant oncology research target[1][2][5][7].
