Vasorin (VASN) is a transmembrane glycoprotein that can be cleaved and released into a soluble form (sVASN). It is a known regulator of the TGF-β signaling pathway, often restraining its proinflammatory fibrotic signaling. Vasorin plays diverse roles in various physiological and pathological processes, including angiogenesis, inflammation, and tumor progression. It has been identified as a potential biomarker and therapeutic target in conditions such as cancer (e.g., hepatocellular carcinoma, bladder cancer, gastric cancer, small-cell lung cancer, rectal cancer, glioblastoma), preeclampsia, septic shock, retinal vein occlusion, polycystic ovary syndrome, myocardial fibrosis, and bone metabolism. Its functions involve interactions with proteins like NOTCH1 and CD71, and modulation of pathways such as YAP/TAZ, PTEN/AKT, and glycolysis.
