Overview

Vatelizumab (formerly GBR 500 or SAR339658) is a humanized monoclonal antibody that targets the alpha-2 subunit (CD49b) of the VLA-2 (very late antigen-2) integrin. VLA-2 is a collagen receptor expressed on various cells, including activated T cells, and plays a role in cell adhesion and migration. By blocking the interaction between VLA-2 and collagen, vatelizumab aims to modulate inflammatory responses. It was originally developed by Glenmark Pharmaceuticals and later licensed to Sanofi. Clinical development has primarily focused on autoimmune and inflammatory conditions such as relapsing-remitting multiple sclerosis (RRMS), ulcerative colitis, and Crohn's disease. Additionally, research has explored its potential in oncology, specifically for enhancing the efficacy of PD-L1 blockade in collagen-rich tumors like breast cancer by promoting CD8+ T-cell immunity.
