**VAX014 + pembrolizumab** is an investigational combination therapy for cancer comprising VAX014, a novel, oncolytic immunotherapy based on targeted bacterial minicells, and pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-1. VAX014 selectively binds and enters tumor cells by targeting α3β1 and α5β1 integrins, releasing perfringolysin O to induce tumor cell lysis and stimulate antitumor immune responses. Pembrolizumab blocks PD-1 receptor-mediated immune evasion by tumors, further enhancing T-cell-mediated antitumor activity. The combination is being evaluated in clinical trials as intratumoral VAX014 injections with systemic pembrolizumab in patients with advanced solid tumors, including those refractory to prior PD-1 blockade[2][4][5].
