Overview

VAX102 is a recombinant fusion protein vaccine candidate developed to enhance immunity against influenza A. It consists of four copies of the ectodomain of the influenza virus matrix protein 2 (M2e) antigen genetically fused to *Salmonella typhimurium* flagellin, which acts as a Toll-like receptor 5 (TLR5) ligand and adjuvant[3][4][1]. The primary mechanism involves stimulating both innate and adaptive immune responses by targeting TLR5 and inducing antibodies against the conserved M2e region of influenza A viruses. This approach aims to provide broad cross-protection against various strains of influenza A not covered by seasonal vaccines[1][4]. Clinical studies have shown that when administered with trivalent inactivated influenza vaccine (TIV), VAX102 can increase immune responses to TIV's H1 and H3 antigens and induce significant anti-M2e antibody production[1][4]. However, higher doses have been associated with increased local and systemic adverse reactions[5].
