VAX128C is a recombinant H1N1 influenza vaccine candidate that consists of the globular head (HA1–2 subunit) of the hemagglutinin protein from A/California/07/2009 (H1N1) fused to two sites on bacterial flagellin (STF2): both at the C-terminus and replacing the D3 domain. This design targets Toll-like receptor 5 (TLR5), acting as both an antigen and self-adjuvant. VAX128C is administered intramuscularly, was developed by VaxInnate, and has undergone Phase 1 clinical trials for influenza vaccination. VAX128C was studied as a single antigen and as the H1N1 component of a quadrivalent influenza vaccine (VAX2012Q), demonstrating good immunogenicity and tolerability in adults.[1][3][5]
