Vaxfectin-formulated H5N1 DNA vaccine is an investigational vaccine candidate developed by Vical Incorporated for the prevention of infection by the H5N1 subtype of Influenza A virus (avian influenza). The vaccine utilizes a DNA plasmid (VCL-A01) encoding the hemagglutinin (HA) protein of the H5N1 virus, which, upon intramuscular injection, is taken up by host cells to induce the expression of the viral antigen. This process triggers both humoral and cellular immune responses, including the production of neutralizing antibodies and the activation of T cells. To enhance immunogenicity, the plasmid is formulated with Vaxfectin, a proprietary cationic lipid-based adjuvant system consisting of GAP-DMORIE and DPyPE. Vaxfectin facilitates the delivery of the DNA into cells and acts as a potent adjuvant to boost the magnitude and breadth of the immune response. The vaccine reached Phase 2 clinical trials, demonstrating safety and the ability to elicit protective antibody titers in healthy volunteers.
