Overview

Vaxonella is an oral vaccine delivery platform developed by Prokarium that utilizes live, attenuated, genetically modified *Salmonella enterica* serovar Typhi bacteria. The platform is engineered to be safe and serves as a vehicle to deliver recombinant protein vaccines directly to the immune system via the gut lining. It incorporates Emergent BioSolutions' Typhella and spi-VEC technologies alongside Prokarium's proprietary ORT-VAC (Operator-Repressor Titration for Vaccines) plasmid stabilization system, which ensures the maintenance of self-replicating DNA plasmids without the need for antibiotic resistance markers. Vaxonella is designed for high thermostability, remaining stable at 40°C for up to 12 weeks and maintaining a shelf life of over three years at 4°C, which facilitates distribution in regions without reliable cold chain infrastructure. It is being applied to develop vaccines for enteric fever (typhoid and paratyphoid), *Chlamydia*, *Clostridioides difficile*, and other infectious diseases.
