VB10.16 is an investigational, off-the-shelf DNA-based therapeutic cancer vaccine developed to treat cancers associated with human papillomavirus type 16 (HPV16), including cervical cancer and head and neck squamous cell carcinoma (HNSCC). The vaccine encodes a recombinant fusion protein consisting of mutation-inactivated HPV16 E6 and E7 oncoprotein antigens, a dimerization module derived from human immunoglobulin G3 (IgG3), and the chemokine ligand CCL3L1, which targets antigen-presenting cells (APCs). This design ensures efficient delivery of HPV antigens to APCs, leading to robust activation of T cells that can recognize and kill HPV-positive tumor cells. Clinical studies have shown that VB10.16 induces strong HPV-specific T-cell responses correlated with clinical efficacy, especially in combination with immune checkpoint inhibitors such as atezolizumab or pembrolizumab[1][4][5][8].
