VB10.2129 is a second-generation prophylactic DNA vaccine candidate developed by Nykode Therapeutics for the prevention of COVID-19. It encodes the receptor binding domain (RBD) derived from the B.1.351 (Beta) variant of concern of SARS-CoV-2 and is designed to induce both antibody and T cell responses against current and future variants of SARS-CoV-2[1][2][5]. Preclinical studies demonstrated that VB10.2129 induces strong cross-neutralizing antibodies against several variants of concern, as well as robust RBD-specific T cell immunity[2][5]. The vaccine protein is expressed and secreted following administration, leading to immune activation[2]. As a DNA vaccine, it works by delivering plasmid DNA encoding the viral antigen into host cells, which then express the antigen to stimulate an immune response[8].
