VBC117 is a first-in-class bispecific antibody-drug conjugate (ADC) that simultaneously targets the Epidermal Growth Factor Receptor (EGFR) and Cadherin-17 (CDH17). Developed by VelaVigo Bio, this therapeutic candidate is designed to treat advanced malignant solid tumors by selectively delivering a cytotoxic payload to cancer cells that co-express both antigens. EGFR is a well-validated oncogenic driver in various cancers, while CDH17 is a cell adhesion protein specifically overexpressed in gastrointestinal malignancies such as colorectal and gastric cancers. By utilizing a bispecific format, VBC117 aims to enhance tumor-specific binding and internalization, potentially reducing the systemic toxicities often associated with monospecific EGFR-targeted therapies. The drug is currently undergoing evaluation in a Phase 1/2a first-in-human clinical trial to assess its safety, tolerability, and preliminary efficacy in patients with refractory solid tumors.
